Modified-release formulation strategies
The increasing popularity of modified-release (MR) dosage forms is attributed to the numerous therapeutic advantages they provide for both drug developers and patients. Oral MR formulations are specifically designed to regulate the rate and/or location of drug release within the gastrointestinal (GI) tract. Unlike immediate-release (IR) formulations, MR dosage forms present several benefits, including:
- Maintaining drug plasma concentrations for an extended period, thereby reducing the frequency of dosing.
- Mitigating drug peak-to-trough ratios, leading to a decrease in peak-related adverse events (AEs) and an enhancement in efficacy.
- Enabling targeted drug delivery to specific regions of the GI tract for localized treatment.
