The tabletability flip phenomenon (TFP) occurs when a solid form of an active pharmaceutical ingredient (API) with lower intrinsic tabletability demonstrates improved tabletability upon blending with identical excipients. This phenomenon emphasizes the crucial role of solid-form selection and formulation design in tablet development. While prior research has explored the mechanisms and factors driving TFP, a dependable approach to predict its occurrence early in formulation design is still required to fully support a quality-by-design framework.
In this study, we evaluated the potential of a recently introduced mixture tabletability model to forecast TFP in two distinct formulation systems. The model successfully anticipated whether TFP would occur in both cases, suggesting its promise as a predictive and diagnostic tool during formulation development. However, the differences observed between predicted and experimental results indicate that particle size may influence the model’s accuracy, warranting further investigation.
